What Is NVG-291?
NVG-291 is a new medicine. It is being tested to help people with nerve damage. Your nerves are like electrical wires in your body. They carry messages from your brain to your muscles. When nerves get damaged, those messages stop getting through. This can happen in spinal cord injuries, Alzheimer's disease, and multiple sclerosis (MS).
NVG-291 is not a pill. It is a peptide. You can think of a peptide as a tiny protein. It is injected under the skin, like an insulin shot.
The company making it is called NervGen Pharma. They are based in Canada. Their stock trades on the Nasdaq under the symbol NGEN.
How Does It Work?
Imagine your nerves have a parking brake. When you are a baby, the brake is off. Your nerves can grow and repair themselves easily. That is how a baby learns to walk.
As you grow up, your body pulls the parking brake on. The brake is a protein called PTPσ (pronounced P-T-P-sigma). It tells your nerves: "Stop growing. We are done here." That brake is useful. It stops nerves from growing where they should not.
But when you get a spinal cord injury, or when you get a disease like Alzheimer's, you WANT those nerves to grow and repair. The brake is now a problem. It stops the healing.
NVG-291 releases the parking brake. It blocks PTPσ. With the brake off, the nerves can start repairing themselves again. This is a brand new idea. No other drug works this way.
Who Is It For?
Right now, NVG-291 is being tested for three conditions:
- Spinal cord injury. This is the most advanced testing. Phase 2 is complete. People with chronic spinal cord injuries (injuries that happened years ago) showed improvements in walking and arm function.
- Multiple sclerosis (MS). MS damages the protective coating around nerves. NVG-291 may help repair that coating.
- Alzheimer's disease. Alzheimer's damages nerve connections in the brain. NVG-291 may help rebuild those connections.

Why Does This Matter?
NVG-291 matters for two big reasons.
First, it is a completely new approach. Most drugs for nerve damage try to protect nerves from dying. NVG-291 does something different. It tells the surviving nerves to grow and reconnect. It unlocks a repair system that is already inside your body.
Second, it could help with many age-related diseases. As we age, our nerves lose the ability to repair themselves. That is part of why our memory gets worse. Why our balance gets shaky. Why we recover more slowly from injuries. A drug that flips the repair switch back on could change how we age.
If NVG-291 works in Phase 3, it would be the first drug to actually repair nerve damage, not just slow down the damage. That would be a historic moment in medicine.
What We Do Not Know Yet
- Phase 3 could fail. Many drugs that look great in Phase 2 fail in Phase 3. The sample sizes are bigger. The bar is higher.
- Side effects are not fully known. The parking brake exists for a reason. Turning it off everywhere in the body might cause problems we do not see in small trials. Unwanted nerve growth is a real risk.
- It may not work for Alzheimer's. The Alzheimer's studies are very early. The brain is much more complicated than the spinal cord.
- Timeline is long. Phase 3 trials take years. Even if everything goes perfectly, NVG-291 is still several years away from FDA approval.
- NervGen is a small company. They may need more funding to finish Phase 3. They may partner with a larger drug company.
The Bottom Line
NVG-291 is the first drug designed to release the brakes on your body's own nerve repair system. Early results in spinal cord injury are encouraging. Phase 3 testing is being planned. If it works, it could change how we treat not just spinal cord injuries, but also Alzheimer's, MS, and age-related nerve decline. But it is still years away from approval, and Phase 3 is where most drugs fail. Watch this space.
